PAPANICOLAOU STAIN
Intended use:
Study of morphology of squamous epithelial cell
Principle:
Papanicolau Stains are used in conjunction with Hematoxylinnuclear stains in the diagnosis of malignant cytological disease.EA-36 and EA-50 are used in conjunction with OG-6 forgynecological staining. EA-65 is used with OG-6 fornon-gynecological staining. When performed properly, the stainedspecimen should display hues from the entire spectrum: red,orange, yellow, green, blue, and violet. The chromatin patternsare well visible, the cells from borderline lesions are easier tointerpret and the photomicrographs are better. The stainingresults in very transparent cells, so even thicker specimens withoverlapping cells can be interpreted.

Physical Appearance:
Hematoxylin clear wine red coloured liquid without anyparticles.
OG-6 clear orange color liquid without any particles.
Light green SF green color clear liquid with reddish purple tingewithout any particles.
Dehydrant colorless liquid without any particles.
Xylene colorless liquid without any particles.
Ana fix colorless liquid without any particles.

PROCEDURE:
1. Prepare a smear on clear, dry glass slide.
2. Fix with Ana fix.
3. Pour Dehydrant Reagent on slide for 3 minutes.
4. Wash with distilled water.
5. Flood the smear with Hematoxylin reagent for 1 min.
6. Add 3 drops of Phosphate Buffer which is not provided for 10seconds.
7. Wash with Dehydrant 2 times for 30 seconds.
8. Pour Working Stain on slide. Allow it to remain for 45seconds.
9. Wash with Dehydrant for 30 seconds.
10. Rinse with Xylene.
11. Allow the slide to air dry.
12. Mount with glass cover using a drop of DPX.
